FAM83E Inhibitors

Chemicals that modulate FAM83E activities can be broadly classified based on the pathways they target: PI3K/AKT and MAPK among them. PI3K inhibitors such as LY294002 and Wortmannin directly interfere with the PI3K/AKT pathway by inhibiting the PI3K kinase. The immediate effect is a decrease in AKT phosphorylation, which is instrumental in multiple cellular activities that FAM83E is implicated in. Notably, NVP-BEZ235 extends its inhibitory effects to mTOR, thereby covering a broader spectrum of intracellular signaling that includes, but is not limited to, FAM83E. MEK inhibitors like U0126 and AZD6244, as well as p38 and JNK inhibitors like SB203580 and SP600125 respectively, affect the MAPK pathway. These inhibitors modulate the activation and functioning of MAPKs, effectively altering cellular activities that FAM83E may influence. Src kinase inhibitors such as PP2 directly target the Src-family kinases, which are implicated in a variety of cellular activities including growth signaling and cellular adhesion. By inhibiting these kinases, PP2 can modulate signaling pathways that may interact with or influence FAM83E's functionalities. In the case of BAY 43-9006, its ability to target both Raf kinase and VEGFR indicates a more complex modulatory effect on the cellular environment where FAM83E operates.
