FAM120C activators work through a diverse array of biochemical mechanisms to enhance the functional activity of this protein. Compounds that raise intracellular cAMP levels act as one category of activators, doing so by stimulating adenylyl cyclase or engaging beta-adrenergic receptors. The resultant elevation in cAMP triggers a cascade that culminates in the activation of protein kinase A (PKA). Once active, PKA can phosphorylate various substrates, including proteins that may interact with or modify the activity of FAM120C, thereby increasing its functional activity within the cell. Similarly, the application of cAMP analogs can directly activate PKA, bypassing upstream receptors and achieving the same end of enhancing FAM120C activity through phosphorylation events. Another pathway implicated in the regulation of FAM120C involves calcium signaling. Agents that elevate intracellular calcium concentrations can activate calcium-dependent kinases, which, in turn, may phosphorylate FAM120C or its regulatory proteins, leading to its enhanced activity.
Additionally, the activation of protein kinase C (PKC) by specific activators can initiate phosphorylation cascades that, either directly or indirectly, result in the activation of FAM120C. Certain activators disrupt normal cellular processes, such as protein synthesis, leading to the activation of stress-activated protein kinases that may also impinge on FAM120C activity. Inhibitors of protein phosphatases, by preventing the dephosphorylation of proteins, can cause a net increase in protein phosphorylation, including that of proteins in pathways associated with FAM120C. Moreover, oxidative stress inducers, such as hydrogen peroxide, can activate multiple kinase pathways, potentially culminating in the activation of FAM120C. Lastly, the modulation of PKC activity through specific inhibitors can lead to compensatory mechanisms in the cellular signaling network, which may inadvertently upregulate pathways that activate FAM120C.
