EF-Ts Inhibitors

EF-Ts inhibitors primarily work by binding to the bacterial ribosome, either to its 30S or 50S subunit, and interfering with various stages of the protein synthesis process, including initiation, elongation, and termination. By doing so, they prevent the proper functioning of the elongation factors, among which EF-Ts is a key player.

EF-Ts inhibitors interfere with the association of aminoacyl-tRNA with the ribosome, which is a step where EF-Ts indirectly plays a role by recycling EF-Tu for subsequent rounds of elongation. They also work by blocking the peptidyl transferase reaction and initiation complex formation, respectively, both of which are essential for continuous protein synthesis and indirectly involve EF-Ts function. These inhibtors also target the translocation step and EF-G cycling, which, while not directly impinging on EF-Ts, would still result in a downstream impact on its function due to the halt in protein synthesis.
