AGRP Activators

Agouti-related protein (AGRP) is a neuropeptide predominantly expressed in the hypothalamus, known for its pivotal role in regulating food intake and energy homeostasis. AGRP acts as an endogenous antagonist of the melanocortin receptors, specifically MC3R and MC4R, which are involved in the control of feeding behavior and energy expenditure. By binding to these receptors, AGRP inhibits the downstream signaling cascade mediated by melanocortin peptides, such as α-melanocyte-stimulating hormone (α-MSH), resulting in increased food intake and decreased energy expenditure.

The activation of AGRP involves complex regulatory mechanisms within the central nervous system. AGRP neurons receive inputs from various neurotransmitter systems, including glutamatergic, GABAergic, and serotonergic inputs, which integrate signals from peripheral metabolic cues such as leptin and ghrelin. Under conditions of energy deficit or negative energy balance, AGRP neurons are activated, leading to increased AGRP expression and release. This activation serves as a homeostatic response to restore energy balance by promoting feeding behavior and reducing energy expenditure. Additionally, AGRP neurons are regulated by a network of upstream hypothalamic nuclei, including the arcuate nucleus and the paraventricular nucleus, which integrate signals related to hunger, satiety, and energy availability. Overall, the activation of AGRP represents a crucial component of the intricate neural circuitry governing energy homeostasis and feeding behavior in mammals.
