ACOT6 Activators

ACOT6 activators employ diverse biochemical mechanisms to augment the enzyme's activity in lipid metabolism. Certain activators enhance intracellular cyclic AMP (cAMP) levels, which in turn activates protein kinase A. This kinase phosphorylates transcription factors that elevate ACOT6 expression and its enzymatic activity. In addition, the intracellular concentration of ACOT6 substrates is increased by other activators, which directly stimulates ACOT6 by providing more molecules for hydrolysis. This leads to the allosteric activation of ACOT6, enhancing its capacity to catalyze the breakdown of fatty acyl-CoA thioesters.

Additionally, activators that inhibit enzymes responsible for fatty acid transport into mitochondria lead to an accumulation of acyl-CoAs, which in turn indirectly increases the activity of ACOT6 as it works to hydrolyze these substrates. Moreover, specific compounds known to elevate NAD+ levels can activate sirtuins, which may deacetylate and activate transcription factors that upregulate ACOT6. Agonists of peroxisome proliferator-activated receptors further contribute to the transcriptional upregulation of ACOT6, leading to an increase in enzyme expression and subsequent rise in its metabolic activity.
