U-46619, a prostaglandin H2 (sc-201266) analogue, has been shown to be a potent agonist of TXA2R (thromboxane A2 receptors) and also can cause constriction of coronary arteries. Thromboxane A2 has been reported to inhibit platelet-rich plasma aggregation. Mechanistic studies suggest that this compound binds to the putative human platelet TXA2R/PGH2 receptor. Once bound, U-46619 activates TXA2Rs and inhibits the release of hippocampal noradrenaline. Induces platelet aggregation at 32nM and activates p38 (p38 MAPK). U-46619 is an activator of ERK 1 and ERK 2.
