Are there supplied data that measured expression in mouse liver? When I check the expression of SQSTM1 at mouse liver, SQSTM1 band is formed at about 55kda. I think it is not 62kda. So I want to see supplied data in mouse liver if you have.

Asked by: dudtmd159
Thank you for your question. The expected molecular weight for SQSTM1 is closer to 62kDa, but there are many reasons why it can run slightly lower. Please contact your local distributor or technical service directly for assistance regarding your WB results, we are happy to discuss the molecular weight for the SQSTM1 protein with you.

Do you have any suggestions for the dilution and incubation time for use of SQSTM1 Ab (D-3) in IHC on paraffin embedded tissues?

Asked by: MGab
Thank you for your question. We recommend a starting dilution of 1:50 for IHC with this antibody. For incubation we recommend starting with a 1 hour incubation, you can do overnight if needed. You can find our protocol here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Which antigen retrival for IHC of formalin fixed, paraffin-embedded tissues do you recommend to use together with this Ab? Which one was used for Immunoperoxidase staining in your case (figure up)?

Asked by: Pauli
Thank you for your question. We do recommend antigen retrieval with this antibody. The recommended method is sodium citrate with heat, this was used for our in house data. You can find the recommended protocol here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Is it possible to use SQSTM1 Antibody (D-3) for double or triple staining tissue samples when my other primary antibodies are also raised in mice?

Asked by: Trav11
When trying to perform double or triple staining of a tissue sample while using multiple primary antibodies raised in mice, we recommend using our directly conjugated mouse monoclonal primary antibodies. Currently, SQSTM1 Antibody (D-3) is available conjugated to HRP (sc-28359 HRP), phycoerythrin (sc-28359 PE), fluoroscein (sc-28359 FITC), Alexa Fluor® 488 (sc-28359 AF488) or Alexa Fluor® 647 (sc-28359 AF647).

What secondary reagent should I use for detection of SQSTM1 Antibody (D-3) in Western blot?

Asked by: Trav11
We recommend using one of our exclusive Mouse IgG Binding Proteins as a secondary detection reagent, such as m-IgGκ BP-HRP: sc-516102, for Western blot. A complete list of all available binding proteins is available on our website here: https://www.scbt.com/scbt/browse/support-products-mouse-igg-binding-proteins/_/N-ecrety

Rated 4 out of 5 by from Good Ab for WB but not for IF I used this Ab for WB and it worked perfectly. However, this Ab did not work properly in IF. It stained nuclei instead of the p62 in cytoplasm.

Rated 4 out of 5 by from works okay I use this antibody in the Hinck Lab for WB and IF. It works well, and I see no background ever, but the A6 lot seems to perform better for my assays. I have had success using both and recommend both.
