PKA Inhibitor IV is a competitive, non-reversible peptide inhibitor for cAMP-dependent protein kinase (PKA). it is a selective inhibitor (Ki ~2-8 nM) of PKA over other kinases; used at 500 nM to improve specificity of other kinase assays. Sequence is derived from the heat-stable skeletal muscle inhibitor protein of PKA, mapping within the N-terminal domain of the cAMP protein kinase inhibitor. The PKA Inhibitor peptide binds to the catalytic subunit of PKA and displaces the regulatory subunit, and mimics protein substrate by binding to the catalytic site via the Arg-cluster basic residues.

Usage :
• Dissolve peptide in distilled water first. The concentration should be high enough to allow for the addition of buffer after the product is dissolved completely.

• If necessary, a few drops of dilute acetic acid or ammonium hydroxide can be added to help solubilize the peptide.

• The solution can be sonicated briefly to help solubility. For storage, solutions should be in the pH range of 5-7.0, aliquoted and stored at -20°C or lower. Solutions can be stored for approximately 3 to 4 months.

(PubMed ID 15044030) used product sc-3010, PKA Inhibitor IV) to ascertain if inhibition of possible K252a sensitive kinases are involved in the survival activity of MC14.
