"preferring positively charged amino acids directly neighbored to phosphoserine". Does it mean that phosphoserine without neighboring positively charged amino acids will not be recognized by the 16B4 Ab ?

Thank you for your question. The antibody recognizes a broad range of serine-phosphorylated proteins in crude cell extracts, interacting with pSK and pSP motifs strongly, recognizing substrates of MAP/SAP kinases and CDC kinases. Phosphoserine detection using an antibody generally is affected by the surrounding amino acids sequence, but it is possible that it can detect phosphoserine without neighboring + charged amino acids.

What is the expected molecular weight in Western blot when using p-Ser Antibody (16B4): sc-81514?

Thank you for your question. This antibody is recommended for detection of a broad range of serine-phosphorylated proteins. Therefore, the observed molecular weight is dependent upon which specific phosphorylated protein(s) you are evaluating with this antibody.
