Is this antibody resistant to formalin fixation and periodate oxidation? Any published reference detailing its validation (eg epitope recognized, selectivity against other transporters)? Thks in advance.

Thank you for your question. This antibody, Mdr-1 (D-11): sc-55510, is not resistant to formalin fixation as it has been successfully tested on formalin-fixed paraffin-embedded tissue sections. This data is available on our website. The antibody has been cited in over 48 publications, however we are not aware of any of these publications describing the exact epitope recognized by this clone, or experimental validation being published for cross reactivity with other transporters. Hi We are using the Mdr-1 antibody and we repeatedly see three band (in two different cell lines): 1- 130-200 kDa smear which is supposed to be the Mdr-1 band. 2 and 3- two separated narrow bands between 35-50 kDa. How could you explain that?

Thank you for your question. We do not have any information about the type of lower molecular weight bands you have described when using this Mdr-1 antibody. We would recommend first performing a secondary antibody control, where you incubate your membrane without primary antibody to determine if any of those bands may be a result of non-specific binding of the secondary antibody alone.

What is the recommended secondary reagent to be used with Mdr-1 Antibody (D-11) in Western blot?

Thank you for your question. Mdr-1 Antibody (D-11) is a mouse monoclonal IgG2b with a kappa light chain. Therefore, we would recommend using our mouse IgG kappa binding protein (m-IgGκ BP) conjugated to Horseradish Peroxidase (HRP): sc-516102. We want to order your anti MDR1 mouse antibody. The band which you show at the site looks like a smear between 130-200kDa. How could this be explained? Thank you in advance. Dina

Thank you for your question. Mdr-1 Antibody (D-11): sc-55510 is a mouse monoclonal antibody which recognizes Mdr-1 of human origin by Western blot. The Mdr-1 protein has a predicted molecular weight of 170 kDa, however it has been described as hyperglycosylated and can appear as a diffuse band by Western blot (reference PMID: 12138126). This antibody is very popular and has been cited in over 40 publications, primarily for use in Western blot. A list of these product citations is available on our website for this antibody.
