I would like to detect GST-tagged proteins by WB. Is the Sc-516102 the correct ab and I do not require a secondary ab for ECL-HRP? Thanks Michael

Thank you for your question. We offer a variety of GST-specific primary antibodies. Our GST Antibody (B-14): sc-138 in particular has been featured in several hundreds of publications and is a reliable research tool. For secondary detection with HRP we recommend our secondary binding protein m-IgGκ BP-HRP: sc-516102. Alternatively, the sc-138 HRP direct conjugate can be used. This product does not require secondary detection step, as it is already conjugated with HRP.

If I were to combine something like sc-271164 as my primary andtibody and the m-IgGκ BP-HRP as my secondary (based on the specifics of the antibodies binding) for western blot, what chemiluminescence would you recommend? Would it work with west femto?

Thank you for your question. I would not recommend using two secondaries, i.e. using both HRP conjugated secondary and a chemiluminescence secondary. I would just select a secondary antibody with a fluorochrome. Does this binding protein bind to denatured antibody (ie. boiled and reduced IP eluate), or does it only bind to native antibody.

Thank you for your question. This will recognize the light chain of mouse IgG under reducing conditions (e.g., if you have boiled and reduced a mouse monoclonal IgG primary antibody used in IP).

Thank you for your question. This is not raised in a host animal like a conventional secondary antibody. This is a recombinant protein that specifically binds to only the kappa light chains of mouse IgG and mouse IgM.

Can PBS with 5% normal blocking serum be used as a blocking agent and diluent in the Indirect Immunoperoxidase m-IgGk BP-HRP protocol (as suggested in the other immunoperoxidase protocols) or is the UltraCruz® Blocking Reagent (sc-516214) a requirement.

Thank you for your question. We don’t use, nor recommend using normal animal serum with our mouse kappa/lambda binding proteins. For indirect immunoperoxidase staining, or IHC(P), tissue section blocking, primary antibody dilution and sc-516102 dilution are all done in UltraCruz® Blocking Reagent: sc-516214. Please see our protocol for further details.

I detected non-specific 25 kDa band, when I did western blot using mouse tissues. In the protocol, you suggested using sc-2002, when should I use sc-2002 in western blot?

Thank you for your question. If you are detecting unexpected signals at 25 kDa it is possible that the band stems from endogenous IgG in your sample. Pre-treating your sample with Protein G PLUS-Agarose: sc-2002 can significantly decrease such signals by binding to endogenous Ig, which can then be removed by centrifugation. We bought sc-69879, 365166 and 390196. and they suggest that to be secondar anticor is sc-516102. Is it true or not and seconder anticor is goat or not?
