Which antigen retrieval solution (acidic or basic) does work best for SC-9168 antibody?

Thank you for your question. We recommend using a sodium citrate buffer (pH 6.0) with heat treatment for antigen retrieval with this antibody. What is the human recombinant sample you have shown as the positive control in the datasheet of Insulin Antibody (H-86): sc-9168. Can you please give me the details about it?

Thank you for your question. The Western blot data shown on the product datasheet for this primary antibody, Insulin (H-86): sc-9168, is against a human recombinant fusion protein corresponding to amino acids 22-110 of human insulin. This recombinant protein was developed by Santa Cruz Biotechnology.

I am running human beta cell lysate in a western blot using this antibody- where should I ideally detect the insulin band? 34kDa, 12 kDa or 5.8kDa- and please justify the same please as 5.8 kDa is the MW of insulin

Thank you for your question. It can be very difficult to detect endogenous insulin protein in cell lysates so we recommend you review the literature for recommended protocols. This antibody, sc-9168: Insulin (H-68), has been cited in the literature for use in Western blot many times and you can find those product citations on our website. The expected molecular weight, under reducing conditions is ~ 12 kDa for the unprocessed protein. The insulin A and B chains are both predicted to be ~24 kDa.
