Diethylamine NONOate/AM is an esterase-sensitive and cell permeable, O2-acetoxymethylated diazeniumdiolate that acts as an intracellular NO donor. Diethylamine NONOate/AM is shown to induce apoptosis and to act as an antiproliferative agent in NO-sensitive human leukemia HL-60 and U937 cell lines (IC50 = 8.3 μM and 53 μM, respectively). The compound also suppresses oxidized low-density lipoprotein-induced caspase-3 activation in bovine aortic endothelial cells. Diethylamine NONOate/AM exhibits enhanced stability in physiological buffers. It may also be useful for examining temporally and spatially controlled NO availability.
