Hygromycin B, which is also designated Hydrovetine and D-streptamine, is an aminoglycoside antibiotic substance naturally produced by Streptomyces hygroscopicus, which also shows anthelmintic activity. Hygromycin B has been shown to effectively inhibit translocation of mRNA and tRNAs on the ribosome in both bacteria and eukaryotes and also has a subtle effect on decoding fidelity as it slightly increases aminoacyl tRNA (A-site tRNA) affinity. Hygromycin B is suitable for the selection of cells transfected with Escherichia coli hygromycin resistance gene (hyg or hph), as well as for the maintenance of the Hygromycin phenotype of resistant cells. Consistent with its more specific effect on translocation, hygromycin B exhibits monophasic inhibition of polypeptide synthesis, suggesting that Hygromycin B binds to a single site on the ribosome. Other alternate designations for Hygromycin B include O-6-Amino-6-deoxy-L-glycero-D-galacto-heptopyranosylidene-(1→2-3)-O-β-D-talopyranosyl-(1→5)-2-deoxy-N3-methyl-D-streptamine; Antihelmycin; Hygromix 2.4; Hygromix-8 and Hygrovetin.

Paginas :
• This product is provided as 20 mL of a sterile filtered solution in PBS.

• Appearance: Solution color can vary from light yellow to a dark brown or caramel color. Concentrated Hygromycin solution tends to have a darker appearance.

• The working concentration for the purpose of selection varies with cell type, media, growth conditions and cell metabolic rate. A titration experiment for each cell type should be performed to determine the amount of Hygromycin B needed to kill non-transfected cells.

• Recommended concentrations for the selection of resistant cells is 25-1000 μg/ml. Commonly used concentrations for selection are 200 μg/ml for mammalian cells, 20-200 μg/ml for plant cells & bacteria cells and 200-1000 μg/ml for fungi. Your optimum concentration should be tested experimentally.

• Control cells should die within 5-7 days after addition of the antibiotic allowing colonies of resistant cells to form by 10–14 days.
