Spi-B Antibody (4G5B3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Spi-B protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA). Anti-Spi-B antibody (4G5B3) is available as the non-conjugated format. Spi-B, a member of the Ets transcription factor family, plays a crucial role in regulating gene expression during lymphoid development, particularly in B cells and immature T cells. A winged helix-turn-helix motif known as the Ets domain allows DNA sequence binding, influencing transcription of genes essential for immune function. Spi-B enhances expression of several B cell genes, including those encoding CD72, CD20, and Btk, which are vital for B cell activation and differentiation. While PU.1, another Ets family member, expresses across various hematopoietic cells, Spi-B expression remains more restricted, accumulating during T-lineage commitment and diminishing significantly after beta-selection checkpoint. This specificity underscores Spi-B′s importance in immune system development and function, making Spi-B (4G5B3) monoclonal antibody an invaluable tool for researchers studying lymphocyte biology and related disorders.
