Glucose is the main source of energy for mammalian cells and its entry is mediated by various transporters. This process involves seven facilitative (GLUT-1 to -7) and 2 concentrative glucose transporters (SGLT-1, SGLT-2) and a sensor (SGLT-3). The SGLT family members use the electrochemical gradient of two sodium ions to transport one glucose molecule. The mRNA of SGLTs increase steadily from the fetal period to maturity along with an increase in their functional activity. SGLT-1 is responsible for the uptake of the dietary sugars glucose and galactose from the intestinal lumen, while SGLT-3 is involved in the detection of luminal glucose only. Both the sodium glucose co-transporters SGLT-1 and SGLT-2 are expressed in kidneys. Mutations in the gene encoding SGLT-2 result in familial renal glucosuria (FRG), an isolated disorder of proximal tubular glucose transport, characterized by abnormal urinary glucose excretion in the presence of normal blood glucose levels.

Thank you for your question. We have not tested this mouse monoclonal antibody, SGLT-2 (D-6): sc-393350, for reactivity in rabbit samples. The antibody is raised against amino acids 226-268 mapping within an internal region of SGLT-2 of human origin. Based on sequence analysis, the immunogen sequence does share 91% sequence identity with the published protein sequence for rabbit SGLT2 (see link below as a reference for that sequence). This is a very high degree of sequence identity, and there is a good chance that this antibody would detect rabbit SGLT-2.

I was wondering if this antibody reacts with mouse SGLT-2. They share 86% identity in the amino acid sequence used as a antigen. If not, could you recommend one for IHC/WB?

Thank you for your question. We have not yet tested this mouse monoclonal, SGLT-2 (D-6): sc-393350, for detection of SGLT-2 in mouse samples. Based on the sequence identity with the antigen, it is highly likely that this antibody will also detect the mouse protein. We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.

sc-393350: SGLT-2 (D-6) is a mouse monoclonal antibody that is specific for SGLT-2. It is not expected to cross-react with SGLT-1.
