Ru360, an oxygen-bridged dinuclear ruthenium amine complex, has been noted to selectively and potently inhibit mitochondrial calcium uptake. Exposure of mitochondria to Ru360 in vitro produced no effect on actomyosin ATPase activity, sarcolemmal Na+/Ca2+ exchange, sarcoplasmic reticulum Ca2+ uptake or release, L-type Ca2+ channel current, or cytosolic Ca2+ transients. These results indicate that Ru360 only affects calcium uniporters in the mitochondria. Ru360 does not compete with ATP.
