Datasheets

    • Anti-PPARγ Antibody (E-8) is a mouse monoclonal IgG1 κ PPARγ antibody, cited in 966 publications, provided at 200 µg/ml
    • specific for an epitope mapping between amino acids 486-505 at the C-terminus of PPARγ of human origin (identical to corresponding mouse sequence)
    • Anti-PPAR gamma Antibody (E-8) is recommended for detection of PPARγ1 and PPARγ2 of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA
    • PPAR gamma Antibody (E-8) is available conjugated to agarose for IP; and to either phycoerythrin or FITC for IF, IHC(P) and FCM
    • also available conjugated to Alexa Fluor® 488, Alexa Fluor® 546, Alexa Fluor® 594 or Alexa Fluor® 647 for WB (RGB), IF, IHC(P) and FCM, and for use with RGB fluorescent imaging systems, such as iBright™ FL1000, FluorChem™, Typhoon, Azure and other comparable systems
    • also available conjugated to Alexa Fluor® 680 or Alexa Fluor® 790 for WB (NIR), IF and FCM; for use with Near-Infrared (NIR) detection systems, such as LI-COR®Odyssey®, iBright™ FL1000, FluorChem™, Typhoon, Azure and other comparable systems
    • also available conjugated to biotin for WB, IHC(P) and ELISA; and to either TRITC or Alexa Fluor® 405 for IF, IHC(P) and FCM
    • TransCruz reagent for Gel Supershift and ChIP applications (sc-7273 X, 200 µg/0.1 ml)
    • Contact our Technical Service Department (or your local Distributor) for more information on how to receive a FREE 10 µg sample of PPARγ (E-8): sc-7273.
    • m-IgG Fc BP-HRP (mouse IgG Fc binding protein-HRP) and m-IgG1 BP-HRP (mouse IgG1 binding protein-HRP) are the preferred secondary detection reagents for PPARγ Antibody (E-8) for WB and IHC(P) applications. These reagents are now offered in bundles with PPARγ Antibody (E-8) (see ordering information below).

Does HEK293 cells express this protein or could HEK293 cells be stimulated to express it?

Asked by: yuner
Thank you for your question. Unfortunately we are unable to answer the expression level in HEK293 cells specifically, but this information is probably available in literature, so I would recommend looking in pubmed. We do have data in many cell lines, and we do see PPARγ expression without stimulation in most cases.

The bands we are obtaining in a WB of PAG with this antibody are around 45kDa, really close to the actin band. I am using a gel concentration of 8%. Is this band that I am obtaining PPAR gamma?? Is there any publication that can prove me it is OK?

Asked by: Biozeros
Thank you for your question. Yes, PPARgamma has been reported at several different sizes, ranging down to 43kDa and up to 67kDa. I would suggest not using actin as a loading control when you are working with PPARgamma. GAPDH or Vinculin may be better so as to not be in the same range as your target protein.

hi, what blocking buffer that i should use with this antibody? is it better with BSA or non-fat milk?

Asked by: hsn14
Thank you for your question. For Western blotting with chemiluminescent detection (HRP), we recommend using 5% nonfat milk + TBS + 0.05% Tween-20 (available pre-made as, TBS Blotto A: sc-2333). For Western blotting with near-infrared (NIR) detection, we recommend using our UltraCruz® Blocking Reagent: sc-516214. A link to our recommended protocol for Western blotting is below as a reference. https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
