p-Ser Antibody (16B4) is a mouse monoclonal IgM (kappa light chain) provided at 50 µg/0.5 ml
raised against phosphoserine
recommended for detection of a broad range of serine-phosphorylated proteins in crude cell extracts, preferring positively charged amino acids directly neighbored to phosphoserine of mouse, rat, human and canine origin by WB and IP
See m-IgGκ BP-HRP (mouse IgGκ binding protein-HRP), our highly recommended recombinant alternative to conventional secondary anti-mouse IgG reagents.

Protein kinases catalyze the phosphorylation of serine, threonine or tyrosine residues in target substrates, providing a mechanism of control for myriad cellular signaling pathways. Several families of kinases phosphorylate both serine and threonine residues in target substrates, including the Raf, Rsk, ROCK, PAK, Ak, and PKC families of serine/threonine protein kinases. The modification of proteins by phosphorylation can result in three dimensional changes to the structure of the protein and thereby alter its enzymatic activity or its ability to interact with other proteins. Antibodies targeted to phosphoserine may be used for the characterization of proteins with phosphorylated serine residues, and for the elucidation of cellular pathways involving serine phosphorylation.

The p-Ser Antibody (16B4) antibody is a monoclonal mouse IgM κ (kappa light chain). Still, you recommend the m-IgGκ BP-HRP: sc-516102 as a secondary antibody. But this a IgG-kappa specific ab. Would it still work?
Asked by: PeterPM
Thank you for your question. Yes, m-IgGκ BP-HRP: sc-516102 can detect any antibody that possesses a kappa light chain, including p-Ser Antibody (16B4): sc-81514, other IgM, and IgA antibodies.

Just wondering if your products of sc-81516 and sc-81514 recognize any of the phosphorylated S/T/Y, or they require certain residues beside these phosphorylated residues (like a certain motif).
Asked by: Zhyuhao
Thank you for your question. These antibodies both detect a broad range of serine-phosphorylated proteins in crude cell extracts, preferring positively charged amino acids directly neighbored to phosphoserine. Additional information regarding the detected residues is not available.

"preferring positively charged amino acids directly neighbored to phosphoserine". Does it mean that phosphoserine without neighboring positively charged amino acids will not be recognized by the 16B4 Ab ?
Asked by: Yo14
Thank you for your question. The antibody recognizes a broad range of serine-phosphorylated proteins in crude cell extracts, interacting with pSK and pSP motifs strongly, recognizing substrates of MAP/SAP kinases and CDC kinases. Phosphoserine detection using an antibody generally is affected by the surrounding amino acids sequence, but it is possible that it can detect phosphoserine without neighboring + charged amino acids.

is there any possibility to receive a sample from this antibody? Thanks for your answer.
Asked by: constanze
Thank you for your question. We do not currently offer sample sizes for our p-Ser Antibody (16B4): sc-81514. This is a very popular and well cited antibody. If you purchase the antibody, please know that it is covered by our generous product warranty for one year from the date of purchase. If it does not work in any of the recommended applications, please contact our Technical Service Department and we can provide you with a replacement or refund.

What is the expected molecular weight in Western blot when using p-Ser Antibody (16B4): sc-81514?
Asked by: cjMara
Thank you for your question. This antibody is recommended for detection of a broad range of serine-phosphorylated proteins. Therefore, the observed molecular weight is dependent upon which specific phosphorylated protein(s) you are evaluating with this antibody.
