Manoalide is a potent irreversible inhibitor of PLC (phospholipase C) and PLA2 (phospholipase A2) (isolated from the marine sesterterpenoid, Luffariella variabilis) as well as a calcium channel protein inhibitor. Manoalide has been shown to irreversibly inhibit human group II sPLA2 (synovial fluid PLA2) as well as bee and cobra venom PLA2. This compound inhibits human recombinant sPLA2 selectively over cPLA2. Manoalide inhibits the release of arachidonate in cultured peritoneal macrophages. Its mechanism of action involves covalent modification of lysine residues.
