Thank you for your question.Yes, our mouse IgG kappa binding protein (m-IgGκ BP) conjugated to fluorescein (FITC) can be used as a secondary detection antibody with each of the primary mouse monoclonal IgG (kappa light chain) antibodies from SCBT that you have inquired about. DAPI can be used as a fluorescent nuclear stain and is compatible with any antibodies that are recommended for IF (immunofluorescence). Yes, our m-IgGκ BP-FITC can be used as a secondary detection antibody with antibodies from other suppliers. Specifically, any primary antibodies which are mouse monoclonal IgG or IgM with kappa light chain.

I have a primary antibody from Santa Cruz Technology, unconjugated mouse monoclonal SHIP-1 P1C1 sc-8425.Would this antibody (m-IgGκ BP-FITC: sc-516140) work as a secondary antibody for this primary for immunofluorescence?

Thank you for your question. Yes, this is a suitable secondary for detection of that primary antibody. SHIP-1 Antibody (P1C1) is a mouse monoclonal IgG1 (kappa light chain), so our mouse IgG kappa binding protein will recognize it specifically.

Is this protein (sc516140) suitable for formalin fixed parafin sections?

Our mouse IgG kappa binding protein (m-IgGκ BP) conjugated to fluorescein (FITC): sc-516140 can be used in the same applications as a conventional anti-mouse IgG secondary antibody conjugated to FITC. We recommend using it in immunofluorescence (IF) and Flow Cytometry (FCM) applications. For immunohistochemistry on formalin-fixed, paraffin-embedded tissue sections we would rather recommend performing immunoperoxidase staining with our HRP conjugated mouse IgG kappa binding protein, m-IgGκ BP-HRP: sc-516102. If you have made good experience with FITC-conjugated secondary antibodies on paraffin-sections in the past, you should certainly be able to use sc-516140 as well. What type of blocking buffer can I use with this m-IgGk BP-FITC?

Thank you for your question. We recommend blocking for 30 minutes in UltraCruz® Blocking Reagent (sc-516214) if using our mouse IgG kappa binding protein (m-IgGκ BP) conjugated to fluorescein (FITC). This blocking buffer is an animal-free alternative to sera, BSA or non-fat dry milk. I used it on PFA-fixed mouse retina vibratome slices. It didn't work at 1:100 and 1:60 dilutions. For comparison, I tried an anti-mouse secondary antibody from another company for the same primary antibody, and I did see a signal.
