Ivachtin (2-[4-Methyl-8-(morpholin-4-ylsulfon yl)-1,3-dioxo-1,3-dihydro-2H-pyrrolo[3,4-c]quinolin-2-yl]ethyl acetate) is a potent, cell-permeable, reversible, non-competitive inhibitor of Caspase-3 (IC50 = 23 nM). Also called Caspase-3 Inhibitor VII. It has been shown to have 10-100 fold selectivity over other caspases and higher anti-apoptotic activity than Z-VAD-FMK (sc-3067). Following use of the inhibitor, standard apoptosis assays can then be used to examine treated cells for evidence of apoptosis.
For the fluorometric determination of caspase-3 activity, the following caspase apoptosis detection kit is available from Santa Cruz: Caspase-3 Kit (sc-4263 AK).
