Gangliosides are key players in determining the nature of tetraspanin interactions and are major components of the tetraspanin web. The GD2 gangliosides are sialic acid-containing glycosphingolipids that play a role in signal transduction and cell-cell recognition. Ganglioside GD2 production is controlled by GM2/GD2 Synthase. It is found in all tissues and localizes to the cell surface. Ganglioside GD2 is involved in the onset of apoptosis by dephos-phorylating focal adhesion kinases. Ganglioside GD2 is abundant on neuroblastoma cells and on all tumors originating in the neuroectoderm, such as malignant melanoma, adult T cell leukemia, and certain colon and gastric cancers. This suggests that Ganglioside GD2 may be a good target for immu-notherapy. Additionally, Ganglioside GD2 is found on mesenchymal stromal cells and may be a unique surface marker.

Just wondering whether you have the sequence for the variable region of GD-2 antibody or can you provide the hybridoma for sequencing.
Asked by: Jack cells
Thank you for your question. This antibody was raised against neuroectodermal tumor cells of human origin. The exact epitope has not been epitope mapped. We are not able to provide the hybridoma for sequencing.

From the description of the product, I understand that the 14(G2a) antibody is provided as the kappa light chain of the antibody. How can the kappa light chain recognise the GD2 sugar without the heavy chain? Many thanks
Asked by: DMax
Thank you for the question. The Ganglioside GD2 (14G2a) is a full mouse IgG2a antibody. We characterized the type of light chain because instead of conventional secondary antibodies against full-IgG we instead use a light-chain-specific binding protein which tends to produce cleaner results compared to conventional secondary antibodies. In this case we suggest using m-IgGκappa-specific BP-HRP: sc-516102 for the detection of Ganglioside GD2 (14G2a) sc-53831.

How can this mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?
Asked by: Cweed
When attempting a double or triple stain with mouse monoclonal antibodies, we recommend using a directly conjugated primary antibody. This product, sc-53831, is available conjugated to FITC, AF488, and AF647.
