I received the free sample of DREAM SC-166916 antibody and I got five different spots in Western Blot assay (using astrocytes samples from mice): 32 kDa (should be DREAM), 40 kDa, 48 kDa, 70 kDa and 80 kDa approximately. What could be those spots?

What is the recommended fixation method for immunofluorescence staining with DREAM (A-9): sc-166916 monoclonal antibody?

Asked by: Cweed
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining

we bought this product a few weeks ago. I'd like know that why this antibody detect both size of dream about 34kDa and 20kDa as ur datasheet mention. please let me know the reason that 20kDa size of dream

Asked by: rosalind
Thank you for your question. On our product datasheet for this mouse monoclonal antibody, DREAM (A-9): sc-166916, we list the predicted molecular weight for the DREAM protein as 32 kDa. It appears that your question is referring to the Western blot data published on the product datasheet. This data is not representative of detection of endogenous DREAM protein. The antibody was used to detect DREAM protein in a transfected whole cell lysate. The full-length DREAM protein was observed in the transfected lysate around 32 kDa. The lower molecular weight band has not been characterized, but it is presumed to be an incomplete translation product of the transfected cDNA.

Rated 5 out of 5 by from All is ok We can see that DREAM has been shown to inhibit the transcription of proteins containing DRE-like motifs, including genes encoding the c-Fos protein for the AP-1 transcription factor, suggesting that DREAM may affect the genes of many cells.
