A unique family of cysteine proteases has been described that differs in sequence, structure and substrate specificity from any previously described protease family. This family, Ced-3/caspase-1, is comprised of caspase-1, caspase-2, caspase-3, caspase-4, caspase-6, caspase-7 (also designated Mch3, ICE-LAP3 or CMH-1), caspase-9 and caspase-10. Ced-3/caspase-1 family members function as key components of the apoptotic machinery and act to destroy specific target proteins which are critical to cellular longevity. Poly(ADP-ribose) polymerase plays an integral role in surveying for DNA mutations and double strand breaks. Caspase-3, caspase-7 and caspase-9, but not caspase-1, have been shown to cleave the nuclear protein PARP into an apoptotic fragment. Caspase-6, but not caspase-3, has been shown to cleave the nuclear lamins which are critical to maintaining the integrity of the nuclear envelope and cellular morphology. Caspase-10 has been shown to activate caspase-3 and caspase-7 in response to apoptotic stimuli.

Hello my name is Aidan. I am doing a research project at UT Southwester research center. I am looking into using a western blot and caspase-9 to detect the amount of P10 in the cell but do not know what lysis buffer to use. do you have any recommendation?

Thank you for your question. We do not yet have a specific protocol recommended for detection of the p10 subunit of caspase-9 with this antibody. We use our standard Western blotting protocol for testing this antibody and observe strong and specific signal for the precursor protein. We use our RIPA Lysis buffer. We would recommend checking some of the product citations this antibody for Western blotting, to see if there is any specific recommendation for inducing and detecting the p10 subunit by Western blot.

Can you recommend a positive control to use with caspase-9 p10 (F-7): sc-17784 monoclonal antibody for Western blot?

We recommend Jurkat (sc-2204), K-562 (sc-2203), HCT-116 (sc-364175), Saos-2 (sc-2235), COLO 205 (sc-364177) or HEK 293 (sc-45136) whole cell lysates to use as positive controls as we have great Western blot data showing specific detection of caspase-9 p10 in these cell lines.

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.
