Calbindin D28K Antibody (D-4) is a mouse monoclonal IgG1 kappa light chain antibody targeting the Calbindin D28K protein, also known as CALB1, calbindin 1, D-28K, or vitamin D-dependent calcium-binding protein. Raised against amino acids 35–84 near the N-terminus of human Calbindin D28K, anti-Calbindin D28K antibody (D-4) detects the protein across multiple species, including mouse, rat, bovine, porcine, and human, via western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Calbindin D28K is a critical calcium-binding protein that functions to buffer intracellular calcium levels, modulating calcium signaling pathways essential for neuronal excitability and neurotransmitter release. Located predominantly in the cytoplasm of neurons within the brain, as well as in the intestine and kidney, Calbindin D28K maintains calcium homeostasis vital for proper cellular function and prevention of calcium-induced cytotoxicity. Structurally, Calbindin D28K contains six EF-hand motifs, helix-loop-helix structures, with four active calcium-binding domains, allowing binding of multiple calcium ions simultaneously. Calbindin D28K interacts with various proteins involved in calcium signaling and may influence the activity of calcium channels and the plasma membrane ATPase. Additionally, Calbindin D28K has been implicated in apoptosis and microtubule function, highlighting a multifaceted role in cellular physiology. By enabling precise detection of Calbindin D28K, Calbindin D28K monoclonal antibody (D-4) supports scientific research into calcium-mediated processes and contributes to advances in neurobiology and cell physiology.
