C2 Antibody (E-7) is a high quality monoclonal C2 antibody (also designated C2 antibody) suitable for the detection of the C2 protein of human origin. C2 Antibody (E-7) is available as both the non-conjugated anti-C2 antibody form, as well as multiple conjugated forms of anti-C2 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. The complement component proteins, C2, C3, C4, and C5, are potent anaphylatoxins that are released during complement activation. Binding of these proteins to their respective G protein-coupled receptors induces proinflammatory events such as cellular degranulation, smooth muscle contraction, arachidonic acid metabolism, cytokine release, leukocyte activation, and cellular chemotaxis. C2 deficiency (C2D) is the most common deficiency of the classical complement pathway and is mostly found in patients with autoimmune disease or susceptibility to bacterial infections. The N-terminal extracellular domain 1 of complement C2 receptor inhibitory trispanning, or CRIT, binds to C2 and specifically interacts with the C2a fragment. In doing so, CRIT blocks C2 cleavage and also prevents the classical pathway of C3 convertase formation.

Can it be used to detect pig-derived C2 by WB or ELISA?

Thank you for your question. We have not tested this antibody, C2 (E-7): sc-373809, for detection of C2 in pig samples and so this species is not recommended at this time.
This monoclonal antibody is raised against amino acids 204-503 mapping within an internal region of C2 of human origin. Based on sequence analysis, this is predicted to share 85% sequence identity with a published C2 protein sequence of pig origin (accession number NP_001095285). This is below our threshold to recommend the antibody for use in pig, however the identity is high enough where there is a good chance that the antibody will also recognize the C2 protein in pig.

Can C2 (E-7): sc-373809 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, C2 (E-7): sc-373809 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here:
