Application: Immunofluorescence in (4% PFA fixed) paraffin embedded specimens of rat colon using heat mediated antigen retrieval with TRIS/EDTA buffer (pH:9, 20min, 92°C) or with citrate buffer (10min, 100°C) and a dilution of 1:50. Conclusion: using the TRIS/EDTA buffer for antigen retrieval, the antibody labels with an excellent resolution all the nerve structures as expected: neurons and nerve fibres of the myenteric plexus and submucosal plexus. Using citrate buffer, the label was identical for distribution although less intense to that observed with the antigen retrieval with TRIS/EDTA buffer.
