Aminoglutethimide has been shown to inhibit human placental CYP19 (aromatase) in vitro. In ovine adrenocortical cells, aminoglutethimide has been shown to inhibit ACTH receptor mRNA expression in a time-dependent manner in vitro. It significantly suppresses baseline ACTH-R mRNA expression and steriod secretion in a dose-dependent manner in vitro in the human NCI-h295 adrenocortical carcinoma cell line. In vivo, Aminoglutethimide is known to accelerate its own metabolism and the metabolism of synthetic dexamethasone and glucocorticoid.
