α4a Tubulin CRISPR/Cas9 Knockout (KO) Plasmid (h2) 는 3개의 plasmids를 포함하며 매개의 plasmid는 Cas9 nuclease 와 하나의 α4a Tubulin-특정된 20 nt guide RNA (gRNA) 를 인코딩하며 gRNA는 knockout 효율을 최대한 낮추도록 디자인 되였습니다.
gRNA sequences는 GeCKO (v2) library에서 찾을수 있으며 직접 Cas9 프로틴을 유도하여 유전자 DNA의(%link_11%) 특정된 위치에서 double strand break (DSB)를 완성할수있습니다.
Cas9로 인한 DSB를 포함한 세포를 선발하려면α4a Tubulin HDR Plasmid (h2) (sc-400016-HDR-2)와의 co-transfection을 권장합니다

α4a Tubulin HDR Plasmid (h2) 는 α4a Tubulin CRISPR/Cas9 KO Plasmid (h2)와의 co-transfection을 권장합니다.
α4a Tubulin HDR Plasmid (h2) 는 2-3 plasmids를 포함하며, 매개의 plasmid는 homology-directed DNA repair (HDR) template 는 α4a Tubulin CRISPR/Cas9 KO Plasmid (h2)로 인한 cut site와 대응됩니다.
매 HDR template은 두개의 800 bp homology arms이 디자인되여있어 Cas9-induced double strand DNA break site주변의 genomic DNA 와 결합합니다
매 HDR Plasmid 에는 puromycin resistance gene이 있어 stable knockout (KO) cells을 얻을수있으며 RFP (Red Fluorescent Protein) gene이 있어 형질주입효율을 관찰할수있습니다.
Puromycin resistance와 RFP genes는 두개의 LoxP sites 사이에 위치하여 Cre Vector sc-418923로 추가가공이 가능합니다.

α4a Tubulin Double Nickase Plasmid (h) 는 한쌍의D10A mutated Cas9 nuclease 와 타깃특정 20 nt guide RNA (gRNA)를 인코딩하는 plasmid를 포함하고있으며 gRNA는 대응 CRISPR/Cas9 KO보다 높은 특이성을 띤 knockout gene을 발현하도록 디자인되였습니다.
한쌍의 gRNA는 약 20bp가량 떨어져 있으며 이는 게놈DNA의 Cas9-mediated double nicking을 발생하여 DSB를 모방합니다.
한쌍의 plasmid 중 하나는 selection에 필요한 puromycin-resistance gene을, 다른하나는 형질주입효율을 확인하는 GFP marker를 보유하고있습니다.

α4a Tubulin CRISPR Activation Plasmid (h) 는 synergistic activation mediator (SAM) transcription activation system으로 특정유전자 발현을 upregulate하도록 디자인되였습니다.
α4a Tubulin CRISPR Activation Plasmid (h) consists of three plasmids at a 1:1:1 mass ratio: a plasmid encoding the deactivated Cas9 (dCas9) nuclease (D10A and N863A) fused to the transactivation domain VP64, and a blasticidin resistance gene; a plasmid encoding the MS2-p65-HSF1 fusion protein, and a hygromycin resistance gene; a plasmid encoding a target-specific 20 nt guide RNA fused to two MS2 RNA aptamers, and a puromycin resistance gene
