Benzonase® Nuclease Lyophilized Powder is a solvent-free version of Benzonase® Nuclease (sc-202391). For liquid format stabilized in glycerol, please consider purchase of (sc-202391), Benzonase® Nuclease.

The Benzonase® enzyme is a genetically engineered endonuclease from Serratia marcescens. Benzonase® Nuclease, also called Golden nuclease, is a highly useful endonuclease for removal of nucleic acids from recombinant proteins and protein fragments, and is also used for the reduction of viscosity in protein extracts.

The enzyme completely digests nucleic acids to 5′-monophosphate terminated oligonucleotides 2–5 bases in length. Although the nuclease is capable of cleavage at nearly all positions along a nucleic acid chain, sequence-dependent preferences have been demonstrated. The enzyme prefers GC-rich regions in dsDNA while avoiding d(A)/d(T)-tracts. Benzonase is ideal for a wide variety of applications where complete digestion of nucleic acids is desirable. Digests native or heat-denatured DNA and RNA. This can also be used for the removal of nucleic acid from protein samples.

Unit definition:
One unit of Benzonase Nuclease is defined as the amount of enzyme that causes a ΔA260 of 1.0 in 30 min, which corresponds to complete digestion of 37 μg of DNA. Note that 1 KU = 1000 units.

用法 :
Format: Lyophilized powder


Reconstitution: It is recommended to reconstitute the lyophilized Benzonase® Nuclease in 100 μl (per 25KU) sterile 50% glycerol to a final concentration of 250U/μl. Solubilize for 30 to 60 minutes at room temperature with occasional gentle mixing by hand. Avoid vigorous shaking or vortexing.


Removal of Benzonase® Nuclease: Benzonase® Nuclease contain no "Tag" and used in downstream processing can be removed by various purification methods according to the purification strategy for the target protein.


Reaction buffer: 50 mM Tris-HCl, 1 mM MgCl2, pH 8.0. For dilute solutions, addition of a carrier protein such as 0.1 mg/ml HSA or BSA is recommended to avoid any enzyme loss from surface adsorption.


The activity of any unknown nuclease can be determined from a single measurement by means of the standard curve. The specific activity of Benzonase® Nuclease is 1.3×106 ~ 1.5×106 unit/mg protein.

Lyophilized Benzonase® Nuclease is shipped at Ambient temperature. Upon receipt, store lyophilized product at -20°C, stable for 1 year. Following reconstitution, store at -20°C to -70°C. Avoid repeated freeze-thaw cycles.
