SAS-6 Antibody (G-1) is a mouse monoclonal IgG1 antibody that detects SAS-6 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SAS-6, a 657 amino acid protein encoded by the human gene SAS6, plays a crucial role in the formation of centrioles, which are essential for proper cell division and organization of the mitotic spindle. Anti-SAS-6 antibody (G-1) localizes asymmetrically next to the centriole at the onset of procentriole formation, highlighting its importance in the early stages of centriole biogenesis. Notably, SAS-6 levels fluctuate throughout the cell cycle, being degraded during mitosis and accumulating again at the end of the G1 phase, a process regulated by the anaphase-promoting complex targeting SAS-6 for degradation via the 26S proteasome. The presence of a KEN box in the C-terminus of SAS-6 is critical for this degradation process. Furthermore, elevated levels of SAS-6 can lead to the formation of multiple procentrioles adjacent to a single centriole, which underscores its regulatory role in centriole duplication and the potential implications for cellular organization and division. SAS-6 monoclonal antibody (G-1) serves as an invaluable tool for researchers studying centriole dynamics and related cellular processes.
