Pellino 3 Antibody (B-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Pellino 3 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Pellino 3 (B-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Pellino 3 protein plays a crucial role in the immune response by acting as a scaffolding component within Toll-like receptor (TLR) and Interleukin-1 (IL-1) receptor signaling pathways, facilitating assembly of signaling complexes that include IL-1R-associated kinase 4 (IRAK4) and tumor necrosis factor receptor-associated factor 6 (TRAF6). This interaction is vital for propagating inflammatory signals, as Pellino 3 also engages with TGFβ-activated kinase 1 (TAK1), which is essential for activating downstream mitogen-activated protein kinase pathways. Notably, Pellino 3 promotes translocation of MAPK-activated protein kinase 2 from nucleus to cytoplasm, thereby activating transcription factor CREB in a p38 MAPK-dependent manner. Furthermore, Pellino 3 physically interacts with NFκB-inducing kinase (NIK) in an IL-1-dependent manner, leading to activation of key transcription factors such as c-Jun and Elk-1, which are critical for regulation of genes involved in inflammation and immune responses.
