IWR-1-endo is a potent Wnt signaling inhibitor. Since Wnt protein signaling leads to β-catenin activation, IWR-1-endo then inhibits a cell-based Wnt/β-catenin pathway reporter response. Wnt-induced accumulation of β-catenin is inhibited by IWR-1-endo, and the destruction complex is stabilized as a result. When IWR-1-endo stabilizes Axin-scaffolded destruction complexes, β-catenin phosphorylation is promoted. Additionally, IWR-1-endo promotes endothelial cell specification of cardiac progenitor cells.
