Cox-2 Antibody (D-12) is a mouse monoclonal IgG1 kappa light chain antibody specific for an epitope mapping between amino acids 586-604 at the C-terminus of Cyclooxygenase-2 (Cox-2) of mouse origin. Cox-2 (D-12) antibody reacts with mouse, rat, and human proteins and is suitable for western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), enzyme-linked immunosorbent assay (ELISA), and immunohistochemistry with paraffin-embedded sections (IHCP). Cox-2, also known as Prostaglandin-Endoperoxide Synthase 2 (PTGS2), is an inducible enzyme located primarily in the endoplasmic reticulum and nuclear envelope within cells, which is crucial for prostaglandin biosynthesis. Cox-2′s cellular localization allows efficient interaction with membrane-bound substrates and access to signaling pathways that regulate inflammation and pain responses. Unlike constitutively expressed Cox-1, Cox-2 shows minimal expression under normal physiological conditions but significantly upregulates in response to inflammatory stimuli, cytokines, and growth factors. Such upregulation leads to increased production of prostaglandins, contributing to inflammation, pain, and fever. Cox-2 overexpression has been implicated in various pathological conditions, including chronic inflammation and progression of certain cancers such as colorectal, breast, and lung cancer.
