CD46 Antibody (E4.3) is a mouse monoclonal IgG2a kappa light chain antibody that detects CD46 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). Anti-CD46 antibody (E4.3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD46, also known as membrane cofactor protein (MCP), is a crucial transmembrane glycoprotein that plays a significant role in regulating the complement system, which is vital for immune response and inflammation. CD46 exists as a non-disulfide-linked dimer and is primarily located on the surface of immune cells such as thymocytes, T cells, B cells, monocytes, and granulocytes, as well as on endothelial and epithelial cells. CD46 on these cells is essential for preventing complement-mediated damage to self-tissues, as CD46 acts as a cofactor for the degradation of C3b and C4b by the serine protease factor I. This regulatory function is primarily attributed to the complement control protein repeats (CCPs) within CD46′s structure, particularly CCP1-4, which are critical for activity. Additionally, CD46 serves as a high-affinity receptor for pathogens such as the measles virus and human herpes virus, highlighting CD46′s importance in both immune regulation and viral pathogenesis. Mouse cells express CRRY, a functional ortholog of human decay-accelerating factor (DAF or CD55) and membrane cofactor protein (MCP or CD46), further emphasizing the evolutionary conservation of CD46′s function across species.

Can we get the immunogen?

Asked by: maomaomao
Thank you for your question. This antibody was raised against human PBL and EBV transformed LCL. The exact epitope has not been mapped, so we are unable to provide further information.

Can CD46 (E4.3): sc-7634 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Asked by: Trav11
Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CD46 (E4.3): sc-7634 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
