Calbindin D9K Antibody (D-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects Calbindin D9K of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Calbindin D9K antibody (D-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental needs. Calbindin D9K plays a crucial role in calcium homeostasis, acting as a calcium-binding protein that helps regulate intracellular calcium levels, which is vital for numerous cellular processes, including muscle contraction, neurotransmitter release, and cellular signaling. Calbindin D9K presence in tissues such as cartilage, bone, and specific epithelial cells underscores its importance in maintaining calcium balance and supporting cellular functions. The gene encoding human Calbindin D9K is located on the X chromosome and consists of three exons, with expression being tightly regulated by various factors, including hormonal influences. Calbindin D9K interaction with calcium ions is facilitated by EF-hand motifs, which are essential for function in calcium signaling pathways. Understanding Calbindin D9K role in these processes can provide insights into various physiological and pathological conditions, making Calbindin D9K monoclonal antibody (D-5) an invaluable tool for researchers studying calcium-related functions in human tissues.

does D5 calbindin-d9k antibody cross react with mouse (it is indicated that the antibody detects calbindin-d9k of human origin)

Asked by: christak
Thank you for your question. We have not yet tested this antibody in mouse samples, and so it is not currently recommended for use in that species. There are a couple of product citations reporting use of this specific antibody in mouse samples. Please refer to PMID's: 21850169 and 20068228 for more details.

I am using Calbindin D9K (D-5): sc-74462 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Randy McDonald
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
