ADAMTS16 encodes ADAMTS-16, a secreted zinc-dependent metalloproteinase in the ADAMTS family that proteolytically remodels extracellular matrix (ECM) components and regulates tissue architecture. By modulating protease activity in the pericellular space, ADAMTS-16 influences ECM turnover, cell–matrix signaling, and microenvironmental cues that affect adhesion, migration, and differentiation programs. Altered ADAMTS16 expression or activity has been investigated in contexts where matrix remodeling is dysregulated, including cardiovascular and renal physiology, reproductive biology, and tumor-associated stromal remodeling. As a matrix-associated protease, ADAMTS-16 is commonly studied alongside pathways linked to ECM organization, proteolysis, and cell signaling at the tissue interface.
ADAMTS-16 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the ADAMTS16 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the ADAMTS16 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the ADAMTS16 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish ADAMTS-16 protein expression.
This CRISPR knockout system enables efficient generation of ADAMTS16-deficient cell models for investigation of ADAMTS-16 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
