Chemical activators of CFAP97 domain containing 2 can induce its functional state through various intracellular signaling pathways. This direct activation of PKC by PMA bypasses the need for upstream signaling that would typically result in PKC activation.
Other compounds influence the phosphorylation state of CFAP97 domain containing 2 indirectly by modulating the levels of intracellular calcium or affecting the balance between phosphorylation and dephosphorylation. Ionomycin, for example, increases intracellular calcium levels, which can activate calcium-dependent kinases capable of phosphorylating CFAP97 domain containing 2. Thapsigargin follows a similar route by inhibiting the sarcoplasmic/endoplasmic reticulum Ca2+ ATPase (SERCA), causing an increase in cytosolic calcium concentration, which may also activate these kinases. On the inhibition front, Calyculin A and Okadaic Acid both act as inhibitors of protein phosphatases 1 and 2A (PP1 and PP2A). The inhibition of these phosphatases prevents the dephosphorylation of CFAP97 domain containing 2, thus maintaining it in an activated phosphorylated state. Lastly, Fura-2 AM serves as a calcium indicator that can indirectly contribute to the activation of CFAP97 domain containing 2 by increasing intracellular calcium, which activates calcium-sensitive kinases that target CFAP97 domain containing 2 for phosphorylation.
