Chemical activators of transmembrane protein 238 can induce functional activation through various cellular signaling pathways by influencing specific biochemical processes. Phorbol 12-myristate 13-acetate (PMA), for example, directly activates protein kinase C (PKC), which is a pivotal kinase known to phosphorylate numerous substrates including transmembrane protein 238, leading to its activation. Similarly, Forskolin, by elevating intracellular cyclic AMP (cAMP) levels, activates protein kinase A (PKA), another kinase that phosphorylates and activates transmembrane protein 238. This enhancement of cAMP also occurs upon stimulation with Isoproterenol, which activates adenylyl cyclase via beta-adrenergic receptors to increase cAMP and thereby activate PKA, culminating in the activation of transmembrane protein 238. Ionomycin functions by increasing intracellular calcium levels, leading to the activation of calmodulin-dependent kinases which can then activate transmembrane protein 238. Epidermal Growth Factor (EGF) and Vascular Endothelial Growth Factor (VEGF) both activate receptor tyrosine kinases-EGFR and VEGFR respectively-that trigger downstream MAPK/ERK and PI3K/Akt pathways, resulting in the phosphorylation and consequent activation of transmembrane protein 238. Insulin similarly activates the PI3K/Akt pathway, which is known to influence the activity of a wide range of proteins including transmembrane protein 238. 1,2-Dioctanoyl-sn-glycerol, a diacylglycerol analog, specifically activates PKC which in turn phosphorylates and activates transmembrane protein 238.

Anisomycin engages by activating the c-Jun N-terminal kinase (JNK), which then phosphorylates transmembrane protein 238, leading to its activation. Sodium Fluoride (NaF) activates G-protein-coupled pathways, ultimately affecting kinases that phosphorylate and activate transmembrane protein 238. In oxidative signaling pathways, Hydrogen Peroxide (H2O2) can activate tyrosine kinases that phosphorylate and activate transmembrane protein 238. Additionally, Calcium Ionophore A23187 increases intracellular calcium levels, thus activating kinases that can phosphorylate and activate transmembrane protein 238, illustrating the diverse mechanisms through which chemical activators can regulate the activity of this protein.
