Chemical activators of family with sequence similarity 177 member A2 (Fam177A2) employ various mechanisms to initiate its activation. Phorbol 12-myristate 13-acetate (PMA), a known activator of protein kinase C (PKC), triggers a signaling cascade that leads to the phosphorylation of Fam177A2, a process essential for its activation. Similarly, diacylglycerol analog 1,2-Dioctanoyl-sn-glycerol (DiC8) can activate PKC, which then phosphorylates and activates Fam177A2. Forskolin, by increasing intracellular cyclic AMP (cAMP), activates protein kinase A (PKA), another kinase capable of phosphorylating Fam177A2, thus contributing to its activation. The cAMP analog 8-Bromo-cAMP also activates PKA, which subsequently phosphorylates and activates Fam177A2. The ionophore Ionomycin raises intracellular calcium levels, which activates calmodulin-dependent kinases, leading to the phosphorylation and activation of Fam177A2. Thapsigargin, which inhibits the sarco/endoplasmic reticulum Ca2+-ATPase (SERCA) pump, results in increased intracellular calcium levels, providing another route for Fam177A2 activation via calcium-dependent phosphorylation.
Continuing with the activation mechanisms, Okadaic acid and Calyculin A, both inhibitors of protein phosphatases, lead to an increase in phosphorylation levels of cellular proteins, which may include Fam177A2, hence maintaining it in an active state. S-Nitroso-N-acetylpenicillamine (SNAP) releases nitric oxide, activating guanylyl cyclase and increasing cyclic GMP (cGMP) levels. This elevation in cGMP can activate protein kinase G (PKG), which can phosphorylate Fam177A2. Zinc chloride, providing zinc ions that function as second messengers within the cell, can activate Fam177A2 indirectly through a phosphorylation pathway. Hydrogen peroxide, a reactive oxygen species, serves as a signaling molecule, which can activate Fam177A2 through redox-sensitive signaling pathways involving phosphorylation by various kinases. Lastly, Anisomycin, which activates mitogen-activated protein (MAP) kinase pathways, also contributes to the activation of Fam177A2 as MAP kinases can phosphorylate and activate this protein. Each of these chemicals provides a unique avenue for the activation of Fam177A2, engaging the protein in multiple signaling pathways and ensuring its functional activation within the cellular environment.
