IDE 1 induces definitive endoderm formation in mouse and human embryonic stem cells (ESCs) (EC50 = 125 nM). It is thought to activate the TGF-β signaling pathway. IDE 1 induces Smad2 phosphorylation and increases levels of Nodal expression.
