PHI-1 Antibody (C-11) is a mouse monoclonal IgG2a antibody that detects PHI-1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PHI-1, also known as PPP1R14B, PLCB3N, PNG, or SOM172, is a 147 amino acid coiled-coil protein that plays a crucial role as an inhibitor of protein phosphatase 1 (PP1), a key enzyme involved in various cellular processes. The importance of PHI-1 lies in its ability to regulate myosin light chain dephosphorylation, which is essential for controlling cell migration, adhesion, and retraction. By interacting with MYPT1, a significant regulator that may act as a tumor suppressor, PHI-1 influences Rho-dependent amoeboid cell behavior, thereby impacting metastasis. Notably, PHI-1 is primarily phosphorylated on threonine 57 and an unidentified serine by protein kinase C, leading to a remarkable 50-fold increase in inhibitory activity upon phosphorylation. The gene encoding PHI-1 is located on human chromosome 11q13.1, highlighting its relevance in both normal physiology and potential pathological conditions.
