USP51 encodes a ubiquitin-specific protease that participates in deubiquitination, a key regulatory step controlling protein stability, localization, and signaling output. By reversing ubiquitin conjugation, USP51 can influence proteostasis and the dynamics of multiprotein complexes, with downstream effects on cell-cycle progression, DNA damage responses, and stress-adaptive signaling. Altered activity of deubiquitinases is frequently linked to dysregulated ubiquitin–proteasome system function observed in cancer biology and other disorders involving genome instability or aberrant signaling. USP51 is therefore of interest for dissecting ubiquitin-dependent pathway regulation and identifying context-specific dependencies in human cell models.
USP51 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the USP51 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the USP51 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the USP51 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish USP51 protein expression.
This CRISPR knockout system enables efficient generation of USP51-deficient cell models for investigation of USP51 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
