Thank you for your question. Yes, sc-81779: ABC1 (A00121.01) is recommended for use in IHC with paraffin-embedded sections. Rated 5 von 5 von aus specific staining with high dilution The primary antibody could be further diluted to decrease non specific staining. The tissue was homogenised in RIPA buffer containing 1mM PMSF, 1µM aprotinin, 1µg/ml Pepstatin A, 0.5 µg/ml okadaic acid, 1 mM benzamidine HCl and briefly sonicated. The supernatant was used as the total lysate. The sample was treated with Laemli buffer at 2 different conditions before loading. 1. lysate was incubated in Laemli buffer for 30 min at RT. 2. lysate was heated to 70 C for 10 min. Both yielded similar results. Proteins were transfered onto nitrocellulose membrane using semi dry transfer.
