RNF183 Activators

Lipopolysaccharide, a component of the bacterial cell wall, engages toll-like receptor 4, setting off a cascade that also activates NF-κB, potentially raising RNF183 levels. Proteasome inhibitors like MG-132 cause an increase in cellular proteins due to the inhibition of their degradation, which can indirectly lead to the activation of NF-κB and subsequent upregulation of RNF183. Other anti-inflammatory agents, such as Sodium Salicylate and Sulfasalazine, modulate the NF-κB pathway through the inhibition of IκB kinase, which normally facilitates the activation of NF-κB.

Curcumin and Resveratrol have been found to exert suppressive effects on the NF-κB pathway, yet their modulation might still result in the altered expression of NF-κB-responsive genes, including RNF183. Curcumin achieves this by inhibiting IκB kinase, while Resveratrol modulates the pathway at different levels. Synthetic compounds like Pyrrolidine Dithiocarbamate (PDTC) and Bay 11-7082 target specific points in the NF-κB activation process. PDTC inhibits the oxidative modification of NF-κB, whereas Bay 11-7082 prevents the phosphorylation of IκBα, impeding the release and nuclear translocation of NF-κB, which could indirectly maintain RNF183 expression at elevated levels. Withaferin A and Piceatannol are additional agents that interact with the NF-κB pathway. Withaferin A is known to inhibit the proteasomal degradation of IκBα, and Piceatannol blocks the activation of IκB kinase, both of which are critical steps for NF-κB pathway activation.
