Chemical activators of Olr1431 can engage with the protein through various pathways, predominantly involving the activation of kinases that phosphorylate Olr1431, thereby increasing its functional activity. Phorbol 12-myristate 13-acetate (PMA) and 4-β-Phorbol, for example, activate protein kinase C (PKC), which is known to phosphorylate target proteins. By activating PKC, these chemicals set off a cascade of phosphorylation events that culminate in the activation of Olr1431. Similarly, Forskolin and 8-Bromoadenosine 3',5'-cyclic monophosphate (8-Br-cAMP) raise the levels of cAMP in the cell, which in turn activate protein kinase A (PKA). The activated PKA can then phosphorylate Olr1431, leading to its activation. Dibutyryl cAMP (db-cAMP) operates through a comparable mechanism, sustaining PKA in an active state that promotes the phosphorylation and subsequent activation of Olr1431.

Oleoyl-L-carnitine has an impact on cellular energy states and can activate energy-sensitive kinases, which then phosphorylate and activate Olr1431. Similarly, 1-Oleoyl-2-acetyl-sn-glycerol (OAG), a diacylglycerol (DAG) analog, activates PKC, which can then phosphorylate and activate Olr1431, exemplifying the interconnectedness of cellular signaling pathways and the diverse chemical means by which Olr1431 can be activated.
