Latrophilin-1 Activators

Latrophilin-1 (LPHN1) is a member of the adhesion G protein-coupled receptor (GPCR) family, characterized by a long N-terminal extracellular domain with various protein-protein interaction motifs. This protein is predominantly expressed in the central nervous system and plays a significant role in brain development and synaptic function.

The structure of LPHN1 includes a GPCR autoproteolysis-inducing (GAIN) domain, which suggests that it undergoes autoproteolytic cleavage, a feature unique to adhesion GPCRs. This cleavage is thought to be essential for its function, although the protein remains non-covalently associated after this process. LPHN1 is known for its interaction with α-latrotoxin, a component of black widow spider venom, which binds to it and induces massive neurotransmitter release, leading to toxic effects.Beyond its interaction with α-latrotoxin, LPHN1 is implicated in the regulation of synaptic connectivity and plasticity. It influences neurite outgrowth and may play a role in the formation of neural circuits during development. The receptor is also thought to mediate cell adhesion processes, which are crucial for the correct wiring of neuronal networks.
