CPT1-C Inhibitors

CPT1-C inhibitors comprise chemicals that either directly inhibit CPT1-C or indirectly affect its activity by modulating associated metabolic pathways, specifically the transport and oxidation of long-chain fatty acids. Inhibitors such as Malonyl-CoA bind directly to CPT1-C, disrupting the enzyme from binding to its substrates and thereby inhibiting its action. Many CPT1-C inhibitors, however, exert their effects indirectly. CPT1-C plays a crucial role in the mitochondrial uptake of long-chain fatty acids, a key step in fatty acid oxidation. Therefore, any compound that modulates this process can indirectly affect CPT1-C. For instance, Etomoxir and Perhexiline inhibit CPT1, the enzyme responsible for the transport of long-chain fatty acids into the mitochondria, which indirectly impacts CPT1-C activity by reducing fatty acid uptake. Similarly, Trimetazidine and Ranolazine inhibit the oxidation of long-chain fatty acids, leading to a shift in cardiac energy metabolism from fatty acids to glucose, thereby indirectly affecting CPT1-C. Some CPT1-C inhibitors act by modulating the availability of carnitine, a molecule required for the transport of long-chain fatty acids into the mitochondria. Mildronate inhibits the synthesis of carnitine, while L-Carnitine, in high concentrations, can act as a competitiveinhibitor of CPT1, both indirectly affecting CPT1-C. Other inhibitors like ST1326, Oxfenicine, Tiglic acid, Hibiscus acid, and GSK837149A target CPT1 in general, thus limiting the overall process of fatty acid oxidation and indirectly affecting CPT1-C activity. These inhibitors demonstrate the multifaceted approach to modulating CPT1-C, through both direct inhibition and the manipulation of related metabolic pathways.

Etomoxir is a potent inhibitor of carnitine palmitoyltransferase 1 (CPT1), the enzyme responsible for the transport of long-chain fatty acids into the mitochondria for β-oxidation. Inhibition of CPT1 by Etomoxir reduces fatty acid uptake, thereby indirectly modulating CPT1-C action.
