cathepsin C Activators

Cathepsin C, also known as dipeptidyl peptidase I, is a lysosomal exo-cysteine protease belonging to the cathepsin family. This enzyme is unique in its ability to sequentially remove dipeptides from the free N-termini of protein substrates. Cathepsin C plays a vital role in the activation of several serine proteases within immune cells, particularly in neutrophils, cytotoxic T lymphocytes, and mast cells. By doing so, cathepsin C is pivotal in processes like bacterial killing, antigen processing, and immune cell-mediated cytotoxicity. Its significance is underscored by the fact that mutations in the gene encoding cathepsin C lead to Papillon-Lefèvre syndrome, a condition marked by periodontal disease and skin lesions. Cathepsin C Activators are a group of chemical entities that enhance or stimulate the activity or expression of cathepsin C. These activators might work by binding directly to the enzyme, modulating its active site, or indirectly through pathways that upregulate its expression or disrupt its degradation. An increased activity of cathepsin C, as a result of these activators, augment the activation of serine proteases within immune cells, thereby amplifying their functional responses. The precise molecular mechanisms, binding sites, and regulatory pathways influenced by these activators remain areas of intense scientific inquiry. Understanding the biochemistry and cellular consequences of cathepsin C activation can shed light on its role in various physiological processes and may pave the way for novel strategies in modulating immune responses at the cellular level.
